Restriction Enzyme Digestion and Mapping
Introduction
For experiment three the class worked with restriction enzymes, Ecor1 and Bamhi. Our goal in this lab was to determine the size of the sample DNA. By measuring the base pair size, after using gel electrophoresis we were able to achieve our goal and determine the size of the sample DNA.
Method and Materials
• Gel electrophoresis Unit
• Centrifuge machine
• Pipettes
• Sample DNA
• Restriction enzymes
Gather the materials to make the general restriction enzyme recipe. Make up two tubes of 10x buffer (2ul), restriction enzyme- one with Ecor1, one with Bamhi (1ul), DNA (4ul), and water (13ul) to equal a total of 20ul. Keep the recipe tube on ice as much as possible. Now mix and centrifuge to insure that the contents are all at the bottom of the tube. Then put the tubes on the temperature block at 37 C for 60 minutes. Lastly, add 3ul of buffer.
